Fine cytochemical localization of lysosomal organelles in phagocytic cells of bivalve molluscs. ArchiMer

Bivalve molluscs possess several types of hemolymph cells (hemocytes), involved in internal defense and other physiological functions. Studies in pathological and experimental conditions have shown their micro- and macrophagic ability. However, the mechanisms of intracellular digestion are incompletely known in these organisms. The lysosomal apparatus of hemocytes was therefore investigated in several species by hydrolase localization with the electron microscope. The cytoplasmic granules of granulocytes displayed a strong acid beta -glycerophosphatase activity. Previous observations had shown that these granules may contain phagocytized material. Role of Tamm-Horsfall protein in the binding and in vivo phagocytosis of type 1 fimbriated Escherichia coli by mouse peritoneal macrophages BJMBR

Tamm-Horsfall glycoprotein (THP) contains manno-oligosaccharides that are recognized by type 1 fimbriae (F1) of Escherichia coli. In the present study, we examined the in vivo phagocytic activity of mouse peritoneal macrophages after treatment of bacteria with THP. At low THP concentrations (12.5 µg/ml and 50 µg/ml) no significant difference was observed in the phagocytosis of E. coli F1+. However, at high THP concentrations (500 µg/ml and 1250 µg/ml) we obtained a reduction of bacterial phagocytosis by mouse peritoneal macrophages.

ABSTRACT The present study evaluated the impact that orchiectomy, a routine but painful intervention in bovine husbandry, can cause on pulmonary immunity. To identify whether orchiectomy can impair lung defense, analyses of serum cortisol concentration and of alveolar macrophage and their function (phagocytosis and respiratory burst) were evaluated. Sixteen Holstein bulls (7 mo old, 250±50kg of body weight BW) were divided in two homogeneous groups - the castrated group and the sham group - and the sample were collected on Days -7, 1 and 7 relative to the day of the procedure. Bonamia ostreae is the causative agent of hemocyte disease. It has been detected first during mortalities occured in Franc in 1979. Bonamiosis spread rapidly among oysters farming in France and Europ. Another species, Bonamia sp. has been detected in New Zealand and Australia. These paasites were both included in the "microcell" group. This group contains 2 others parasites, Mikrocytos mackini affecting Crassostrea gigas in British Columbia (Canada) and M. roughleyi affecing Saccostrea glomerata in Australia. The uptake of lipids and lipoprotein particles by macrophages undergoes phagocytic activation and the formation of foam cells are key events in atherosclerosis. In this study we determined how intact high density lipoproteins (HDL) and apolipoproteins-HDL (removal of the lipid component from HDL, i.e., apoHDL) influence the phagocytosis of zymosan by mouse peritoneal macrophages. Zymosan particles preincubated together with lipoproteins or alone (control) were incubated with the macrophages. Farming of the flat oyster Ostrea edulis in Europe is severely constrained by the protozoan Bonamia ostreae. The introduction of the resistant species Crassostrea gigas has been a relief for the farmers, while the pilot programmes to select O. edulis strains resistant to bonamiosis performed in various countries can be seen as a promising strategy to minimise the effects of bonamiosis. However, the physiological bases of this differential susceptibility remain unknown. In the context of Pacific oyster C. gigas summer mortality, the present study demonstrated that the reproductive effort reduced not only the energetic parameters (carbohydrates and CEA) but also the immune parameters (concentration, phagocytosis and adhesive capacity of hemocytes). Increasing food supply from 4% to 12% (of oyster dry weight) did not counteract this phenomena as most of the additional energy was dedicated to the gametogenesis processes. However, after the gametogenesis, the 12% diet clearly favored energy storage restoration and hemocyte production. Hemocyte parameters were also modulated by the food quality. Impact of food availability on energy storage and defense related hemocyte parameters of the Pacific oyster Crassostrea gigas during an experimental reproductive cycle ArchiMer

The aim of this study was to test the effect of food quantity on energy storage and defense capacities of oysters during a reproductive cycle. One-year-old Crassostrea gigas oysters were fed two different dietary rations (4% and 12% of oyster dry weight in algal dry weight per day) in controlled experimental conditions over an annual cycle. Oyster dry weights, carbohydrate and lipid contents, energetic adenylate charge, and hemocyte parameters of oysters were significantly affected by reproductive processes related to seasonal temperature variation and, to a lesser extent, by the dietary rations. The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. Oysters and clams were fed each of three cultured algae: Chaetoceros calcitrans, which is rich in 20:5(n-3) and 20:4(n-6) and poor in 22:6(n-3) fatty acids; T-Iso (Isochrysis sp.), which is rich in 22:6(n-3) and deficient in 20:5(n-3) and 20:4(n-6); and Tetraselmis suecica, which is deficient in 22:6(n-3) and contains only small amounts of 20:5(n-3) and 20:4(n-6). Lactobacillus spp. isolated from different portions of chickens' gastrointestinal tract were evaluated concerning their ability to survive in a water-in-oil (W/0) emulsion containing sesame and sunflower oil. After sixty days of emulsion storage under refrigeration, three of five strains tested survived in number equal to or higher than 10(6)cfu/g. Since 1997, populations of the European abalone Haliotis tuberculata suffer mass mortalities attributed to the bacterium Vibrio harveyi. These mortalities occur at the spawning season, when the abalone immune system is depressed, and when temperatures exceed 17 °C, leading to favorable conditions for V. harveyi proliferation. In order to identify mechanisms of disease resistance, experimental successive infections were carried out on two geographically distinct populations: one that has suffered recurrent mortalities (Saint-Malo) and one that has not been impacted by the disease (Molène). The ultrastructural observation of the hemocytes of D. saccharalis larvae showed different hemocyte tvpes in various stage of interaction with microorganisms, which were interpreted as bacteria. The microorganisms were observed either free in the hemolymph or into the insect hemocytes. The bacteria were detected mostly into lhe granulocytes and plasmatocytes: the spherule cells and the oenocytoids also presented intracytoplasmic microorganisms, but less frequently. The fagocytic capacity of the different hemocyte tvpes is discussed.

The defense reactions against biological (Histoplasma capsulatum and Escherichia coli) and non-biological materials (China ink and nylon thread) were tested in vivo in third instar larvae of Dermatobia hominis. The cellular defense performed by larval hemocytes was observed under electron microscopy. China ink particles were phagocytosed by granular cells 5 h after injection. E. coli cells were internalized by granular cells as early as 5 min after injection and totally cleared 180 min post-injection, when many hemocytes appeared disintegrated and others in process of recovering. The aim of this work was to evaluate the effect of broiler breeders' age and yeast wall (YW) levels on broilers' performance and macrophage activitys. The experiment consisted in a completely randomized design and a 2 X 5 factorial arrangement and two controls of two broiler breeders age (34 and 57 weeks of age) and five YW levels (zero, one, two, three or four kg of YW/ton of diet). The effects of bovine S protein (vitronectin) on phagocytosis of Streptococcus dysgalactiae strains isolated from cattle with mastitis were investigated. Phagocytized streptococci were determined by a fluorometric microassay using glass adherent polymorphonuclear neutrophils (PMN). Preincubation of S. dysgalactiae with bovine S protein significantly increased their phagocytosis by PMN. Bovine S protein had no effect on phagocytic killing of non-S protein binding S. pyogenes cultures. Enzymatic digestion of the bovine S protein binding sites on S. dysgalactiae with pronase resulted in a significative reduction of the effects of S protein on phagocytosis. Shellfish industry is mostly realized in estuary zones, which are subjected to pollutions due to anthropic activities. The harmful effects of pollutants on animals inhabiting these estuarine zones are poorly known. Parameters of hemocyte populations have been considered as relevant indicators of bivalve health and are currently used in immunotoxicological studies. Hemocytes in hemolymph can be collected by puncturing either the pericardial cavity or the adductor muscle sinus with a syringe.
